  • Iodination and Coupling: Iodide is oxidized and attached to tyrosine residues within thyroglobulin to form monoiodotyrosine (MIT) and diiodotyrosine (DIT). MIT and DIT can then undergo coupling reactions to form triiodothyronine (T3) and thyroxine (T4).
  • Hormone Release: When stimulated, the follicular cells engulf the colloid containing the iodinated thyroglobulin, digest it, and release T3 and T4 into the bloodstream.

Thyroid Hormones and Metabolism:

Thyroid hormones, T3 and T4, have profound effects on metabolism:

  • Basal Metabolic Rate (BMR): Thyroid hormones increase BMR, leading to increased energy expenditure and heat production.
  • Carbohydrate and Fat Metabolism: Thyroid hormones enhance the breakdown of carbohydrates and fats, promoting their utilization for energy production.
  • Protein Synthesis: Thyroid hormones stimulate protein synthesis and degradation, influencing tissue growth and maintenance.

Regulation of Thyroid Hormone Secretion:

Thyroid hormone secretion is regulated by a negative feedback loop involving the hypothalamus and pituitary gland:

  • Hypothalamus: The hypothalamus produces thyrotropin-releasing hormone (TRH) in response to low thyroid hormone levels.
  • Pituitary Gland: TRH stimulates the anterior pituitary gland to release thyroid-stimulating hormone (TSH).
  • Thyroid Gland: TSH binds to receptors on the follicular cells, stimulating the synthesis and release of T3 and T4.
  • Feedback Loop: Elevated levels of T3 and T4 inhibit the release of TRH and TSH, thereby reducing thyroid hormone production.

Clinical Significance:

Disorders of the thyroid gland can lead to various conditions:

  • Hyperthyroidism: Excessive production of thyroid hormones results in an overactive thyroid, leading to symptoms such as weight loss, rapid heartbeat, and heat intolerance.
  • Hypothyroidism: Insufficient production of thyroid hormones leads to an underactive thyroid, causing symptoms such as fatigue, weight gain, and cold intolerance.
