What is NLRP12-Associated Autoinflammatory Disorders (Definition/Background Information)

  • NLRP12-Associated Autoinflammatory Disorders (NLRP12-AID) are a group of rare genetic disorders caused by mutations in the NLRP12 gene. This gene plays a role in the body’s inflammatory response, and the mutations result in an overactive immune response, leading to chronic inflammation
  • The main risk factor for NLRP12-Associated Autoinflammatory Disorders is having a family history of the disorder. Most of the cases are manifested in childhood, and these disorders can cause widely varying signs and symptoms including recurrent fever, abdominal pain, skin rashes, joint pain, and recurrent infections

In these types of conditions, the individual has one normal copy and one mutant copy of the gene. The abnormal gene dominates, masking the effects of the correctly functioning gene. If an individual has an autosomal dominant condition, the chance of passing the abnormal gene on to their offspring is 50%. Children, who do not inherit the abnormal gene, will not develop the condition or pass it on to their offspring.
